Scrapie is a fatal transmissible degenerative disease of the central nervous system occurring naturally in sheep and goats, and the prototype of transmissible spongiform encephalopathies (TSE) or prion diseases. The name "Scrapie" was coined as infected animals usually scrape off their wool from itching and rubbing. Although natural transmission of Scrapie to other species has not been shown, TSEs are the subject of increased attention and concern because of the discovery of BSE in cattle and the link between it and variant Creutzfeldt-Jakob disease in humans. Genotyping determines the amino acids coded by five different codons (112, 136, 141, 154, 171) in the prion protein gene (PrP). These are reported as a pair of one character amino acid codes such as RR, RH, QQ, etc. Though there are multiple genes expressing different levels of susceptibility, codon 171 is the gene most focused on in the U.S. At this gene, "QQ" animals are highly susceptible to Scrapie infection if exposed, "QR" are rarely susceptible, and "RR" are highly resistant. This testing does not diagnosis Scrapie, but is an important tool for selecting genetically resistant breeding stock.

Scrapie is a fatal, degenerative disease affecting the central nervous system of sheep and goats, in the same class of diseases as BSE in cattle and Chronic Wasting Disease in elk and deer. Transmission of the classical scrapie agent is not completely understood but apparently healthy sheep infected with the agent can transmit disease. The infecting agent or prion is a mal-formed version (PrPsc ) of a normal protein (PrP) found in all animals. The malformation of the normal prion proteins occurs when they are exposed to the mal-formed version of the protein. The normal prion proteins may be more or less susceptible to malformation when they are exposed to the mal-formed protein based on their genetic makeup. Resistance or susceptibility has been linked to the presence of certain amino acids at various locations within the normal protein. Genotyping determines the amino acids present at specific locations or codons in the prion protein. These are reported as a pair of one character amino acid codes such as RR, RH, QQ, etc. This testing does not diagnosis Scrapie, but is an important tool for selecting genetically resistant animals for breeding and eradication. Codons associated with susceptibility/resistance to scrapie include codon 112, 136, 141, 154 and 171. Codon 171 codes for either arginine ®, glutamine (Q), histidine (H) or lysine (K). Resistance or susceptibility has been associated with several different codons but in the US, codon 171 is believed to be the major determinant of relative scrapie susceptiblity. The presence of R at codon 171 has been associated with increased resistance to scrapie. The presence of Q at codon 171 has been associated with increased susceptibility to scrpaie. The presence of H or K has not been closely associated with resistance or susceptibility and are therefore assumed to be susceptible and equivalent to Q for purposes of the eradication program.
