Whole chromosome painting probes are collections of smaller probes that bind to a different sequence along the length of a given chromosome. Using multiple probes labeled with a mixture of different fluorescent dyes, each chromosome is "painted" using its own unique color, and the resulting full-color map of the chromosome is called a spectral karyotype. Chromosome painting probes are useful for examining complex chromosomal structural aberrations, for example, translocations or subtle rearrangements. Complex chromosome structural alterations can be detected by using cytogenetic methods such as chromosome painting analysis. Spectral karyotyping (SKY) and multiplex (M)-FISH are two types of chromosome painting methods. Both involve "painting" each chromosome a different color using 24 unique fluorochromes and then using the colors to identify subtle structural variations that previously could not be detected. According to one study, M-FISH has been able to identify chromosome abnormalities in up to 7% of patients with previously unexplained mental retardation. Complex structural aberration analysis can be the next step in evaluation when an abnormality is detected but cannot be further identified using more common methods, and the advanced methodologies are being utilized in different domains, including prenatal diagnosis as well as solid and hematologic malignancies. FISH (fluorescence in situ hybridization) is a cytogenetic technique used to detect and localize the presence or absence of specific DNA sequences on chromosomes. FISH uses fluorescent probes that bind to only those parts of the chromosome with which they show a high degree of sequence similarity. Fluorescence microscopy can be used to find out where the fluorescent probe bound to the chromosomes. FISH is often used for finding specific features in DNA for use in genetic counseling, medicine, and species identification. FISH can also be used to detect and localize specific mRNAs within tissue samples. In this context, it can help define the spatial-temporal patterns of gene expression within cells and tissues.
