B cell-activating factor (BAFF) is an immunomodulatory cytokine that advances the survival of B cells, the class switch recombination of heavy-chains, and can also activate T-cells. BAFF-R is the BAFF cell surface receptor and is present on the majority of B cells and a subset of T cells in the circulation. BAFF and BAFF-R normally play a role in B- and T- cell development and regulating humoral immunity, but in pathological conditions, may promote the survival of malignant B cells. The expression of BAFF-R has been associated with 78% of lymphoproliferative disorders including follicular lymphoma, marginal zone lymphoma, chronic lymphocytic leukemia, hairy cell leukemia, and diffuse large B-cell lymphoma, but not others such as precursor B lymphoblastic lymphoma, Burkitt lymphoma, and nodular lymphocyte-predominant Hodgkin lymphoma. In addition, BAFF-R is not a marker for classical Hodgkin lymphoma and T-cell lymphomas. PMID: 16226112

The protein encoded by this gene, transmembrane activator and calcium modulator and cyclophilin ligand interactor (TACI), is a lymphocyte-specific member of the tumor necrosis factor (TNF) receptor superfamily. It interacts with calcium-modulator and cyclophilin ligand (CAML). The protein induces activation of the transcription factors NFAT, AP1, and NF-kappa-B and plays a crucial role in humoral immunity by interacting with a TNF ligand.
