Factor VIII replacement is the foundational treatment for hemophilia A, and the developement of alloantibodies (known as inhibitors) against the replacement factor is the most important complication causing morbidity in hemophilia patients. Antibodies against factor VIII can be inhibitory or noninhibitory, with inhibitors generally being polycloncal IgG directed against the A2, A3 and C2 domains of the factor VIII molecule.
