The appearance of abnormally long or short microsatellites in an individual's DNA is referred to as microsatellite instability. Microsatellite instability (MSI) is a condition manifested by damaged DNA due to defects in the normal DNA repair process. Sections of DNA called microsatellites, which consist of a sequence of repeating units of 1-6 base pairs in length, become unstable and can shorten or lengthen. Microsatellite instability (MSI) is a change in the number of repeats of a specific DNA marker in cancer cells compared to the number of repeats present in non-cancerous cells of the same patient. Different tumor types are associated with different MSI markers, and both the presence of MSI in specific markers as well as the the number of distinct markers demonstrating MSI is important. Tumor cells are classified as being stable, having high instability (MSI-H), or having low instability (MSI-L) based on the number of markers demonstrating MSI. MSI analysis may help guide therapy and be a prognostic indicator. Specific MSI markers include BAT25, BAT26, D2S123, D5S346, D17S2720, NR21, NR24 and MONO27. MSI has been most studied in colorectal cancer, and colorectal tumors that are MSI-H are more likely to be carcinomas than adenomas.
