Neutralization methods such as fluorescent focus neutralization (FFN) is used to confirm positive Porcine Reproductive and Respiratory Syndrome (PRRS) results found with screening methods such as ELISA. It uses sample and control serums that are diluted and incubated on the PRRSV infected cells followed by florescent microscopic examination for characteristic patterns of fluorescent staining. Antibody can be detected in experimenally infected pigs 7 to 10 days post inoculation. Serum samples are screened at a 1:20 dilution to determine the presence or absence of the organism. (USDA).

Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) causes an infection in pigs sometimes known as Blue-Ear Pig Disease. This economically important pandemic disease causes reproductive failure in breeding stock and respiratory tract illness and has shown differences in virulence, tissue trophism and replication capabilities among strains.
PRRSV has two genotypes - the North American strain (type 2 PRRSv, with the isolate prototype VR2332) and the European strain (type 1 PRRSv, with the isolate prototype "Lelystad"). Both types were discovered in their local regions around 1990 but they now share worldwide distributions. These two main genotypes differ genetically by approximately 40%. Within each genotype there is nucleotide divergence of up to 30% (Leung et al., 2010). The majority of the nucleotides that make up the two are the same but diversity within each genotype is high with a significant geographic pattern for the various types. The use of sequence information to compare PRRSV isolates is a valuable tool to monitor PRRSV strains both globally and within a farm over time. It can differentiate between resident and newly introduced viruses as well as between vaccine and field strains.

Neutralizing antibodies are antibodies that interfere with the interaction between the virus and its host cell receptor, therefore preventing infectivity. Neutralizing antibodies can be detected using a traditional VNT or other methods such as pseudovirus VNT and surrogate VNT. VNT and pseudovirus VNT use live virus (the actual virus or a recombinant virus that incorporates the viral antigen of interest, respectively) and cell culture to assess the virus-host interaction and whether neutralizing antibodies are present that block that interaction. Surrogate VNT (sVNT) does not use live virus but rather uses purified viral antigen and host cell receptor protein to mimic the same interaction. The VNT involves incubating patient serum with the virus of interest, and then inoculating onto cell culture. If neutralizing antibodies are present and the virus is neutralized, the cells will not be infected. If neutralizing antibodies are not present, the virus will infect the cells in the cell culture.
