Part Descriptions
LP150045-5 Sequencing
Sequencing is a method used to determine the sequence of individual genes, larger genetic regions (i.e. clusters of genes or operons), full chromosomes or entire genomes. Historically, most sequencing has been performed using the chain termination method developed by Frederick Sanger in 1977. PMID: 271968 Sequencing technologies have improved dramatically, making them cheaper, faster, and more accurate. Next-generation sequencing (NGS), also known as high-throughput sequencing, deep sequencing, and second-generation sequencing, is a type of technology that uses parallel sequencing of multiple small fragments of DNA to determine sequence. This "high-throughput" technology has increased the speed and amount of DNA sequenced at a significantly reduced cost. PMID: 18576944 Several NGS platforms (ie, sequencing instruments and associated reagents) have been developed. Third-generation sequencing is another methodology currently under development that uses parallel sequencing similar to NGS. In contrast to NGS, third-generation sequencing uses single DNA molecules rather than amplified DNA as a template. PMID: 20858600

LP262511-1 Brachyspira sp nox gene
A polymerase chain reaction (PCR) assay using nox-specific restriction fragment length polymorphisms (RFLPs) has been developed that can differentiate all five species of porcine Brachyspira. PMID: 12089283 The nox gene encodes NADH oxidase in Brachyspira species. Species of Brachyspira can be differentiated based on the nox gene sequence due to its variability across species. PMID: 26458507 Several species of the gram-negative spirochete Brachyspira can cause swine dysentery (SD) including B. hyodysenteriae, B. intermedia, B. innocens, B. murdochii, B. aalborgi, B. alvinipulli, B. suanatina, and a provisional species called Brachyspira hampsonii. B. hyodysenteriae, B. suanatina, and B. hampsonii cause a severe mucohaemorrhagic colitis found mainly in grower and finisher pigs. Brachyspira that are weakly heamolytic causing a mild colitis include B. murdochii, B. intermedia, B. aalborgi, and B. alvinipulli in pigs, chickens and humans. Transmission of Brachyspira sp can occur in several ways including: pigs that have subclinical infection; infection from birds or feral animals that act as potential reservoirs; reduced immunity in a herd; routine diagnostic and control measures are not adhered to; and environmental changes such as a change in diet, co-infections and other stresses that increase norepinephrine levels in the gut that can result in the growth and colonization of some species of Brachyspira. Diagnoses of Brachyspira species are primarily made using polymerase chain reaction (PCR) test and/or immune staining.
