The NY-ESO-1 antigen (also referred to as CTAG1) was originally identified in esophageal cancer cells in New York City. It is only found in the developing sperm and is lost as the sperm differentiates. NY-ESO-1 is a member of the cancer-testis gene family which only express antigens in germ cells. NY-ESO-1 is not expressed in normal somatic cell tissue, but is often expressed in cancer cells. Its function is not known due in part because it lacks evolutionary homologs, eliminating the ability to use animal models for research. Because it is rarely present in normal tissue cells, but often present in cancer cells it is a useful biomarker for the detection of early cancer. NY-ESO-1 has been found in one third to one fourth of all melanoma, lung, esophageal, liver, gastric, prostate, ovarian, or bladder cancers. In cancers including colorectal, pancreatic, renal, and lymphomas, NY-ESO-1 is very rarely identified.
