Part Descriptions
LP250692-3 Haemophilus parasuis
Haemophilus parasuis is a gram-negative organism that is harbored in the upper respiratory tract of healthy pigs. It causes Glasser's disease, which is characterized by polyarthritis, polyserositis and meningitis and associated with high morbidity and mortality. Virulence factors include resistance to phagocytosis as well as endothelial invasion, polysaccharide capsule, and biofilm production. Biofilms are enclosed clusters of bacterial cells that are able to resist the host immune system as well as environmental factors such as dessication. PMID: 25428823

Veterinary medicine:
Haemophilus parasuis is a gram-negative bacterium that was first identified in 1992 and infects the upper respiratory tract of pigs. It causes worldwide systemic disease in pigs and is also known as Glässer's disease. Symptoms of severe disease include arthritis, meningitis, polyserositis, septicemia, and pneumonia. H parasuis is the primary cause of death in nursery herds and the third highest in finishing herds in the United States. Knowing the serotype of H parasuis infecting a herd helps to guide vaccination programs and prevent future outbreaks. Fifteen serovars were identified in 1992 using gel immunodiffusion (GID). Indirect hemagglutination (IHA) replaced GID as the test of choice and improved the percent of typable serovars from 60% to 80%. However, IHA has its own issues which include difficulties in obtaining specific antisera, variation in the growth rates between isolates, and very few laboratories perform the assay which is time-consuming, expensive and requires a pure culture. More recently a multiplex polymerase chain reaction (mPCR) assay has been developed that offers rapid and more accurate serovar identification. It uses the highly genetically variable capsule loci of the 15 serovars as the targets. Serovars 5 and 12 are indistinguishable by this method, suggesting that the difference in gene expression of the two is too subtle to detect, or that the serovars are in fact the same and an artifact of IHA typing antibodies. The virulence of the different serovars are reported to differ significantly, and mPCR results are able to accurately distinguish them and determine if multiple serovars are present in a sample. The rapid identification of serovars during an outbreak facilitate the availability of prophylactic vaccination. PMID: 26424843
