This term is created for but not restricted in use to a Nijmegen modification of the classic Bethesda inhibitor titration assay using a chromogenic method. The use of a chromogenic method allows factor VIII inhibitor to be detected in patients receiving emicizumab / Hemlibra®, a therapeutic that interferes with standard clot-based methods PMID: 29446525.

Factor VIII replacement is the foundational treatment for hemophilia A, and the developement of alloantibodies (known as inhibitors) against the replacement factor is the most important complication causing morbidity in hemophilia patients. Antibodies against factor VIII can be inhibitory or noninhibitory, with inhibitors generally being polycloncal IgG directed against the A2, A3 and C2 domains of the factor VIII molecule. The resulting steric hindrance can block functional epitopes including FIX, phospholipid, or von Willebrand binding sites PMID: 23610614. Source: Regenstrief LOINC
