S-Adenosylhomocysteine (SAH) is formed by the demethylation of S-adenosylmethionine (SAMe) and is rapidly metabolized to homocysteine. The reaction of SAH to homocysteine is catalyzed by S-adenosylhomocysteine hydrolase and is reversible with the equilibrium favoring formation of SAH. Therefore, increased levels of SAH may occur when the intracellular concentration of homocysteine increases. Since SAH is known to inhibit methylation dependent reactions, the ratio of SAMe to SAH is often used as an index of methylation status. Disturbances in the transmethylation pathway indicated by abnormal SAMe, SAH or their ratio have been reported in many neurodegenerative diseases, such as dementia, depression or Parkinson's disease. It plays a key role in the maintenance of human health and in the prevention of disease.
