CAGE-1 antibody is one of the several autoantibodies tested to detect various types of cancer in their early stages. The target protein for the CAGE-1 antibody is located mainly in the nucleus of cancer tissues compared to a much lower presence in non-cancerous tissues. The presence of the CAGE-1 protein being found preferentially in cancerous tissue leads to the speculation that it may be associated with tumor growth. The melanoma-associated antigen, or MAGE-A4, belongs to the Cancer-Testis Antigens (CTAs) family. There is some evidence that it may be involved in embryonal development and in tumor transformation or progression. CTAs have been found in several cancers and are able to stimulate antibody and cellular immune responses. The NY-ESO-1 antigen (also referred to as CTAG1) was originally identified in esophageal cancer cells in New York City. It is only found in the developing sperm and is lost as the sperm differentiates. NY-ESO-1 is a member of the cancer-testis gene family which only express antigens in germ cells. NY-ESO-1 is not expressed in normal somatic cell tissue, but is often expressed in cancer cells. Its function is not known due in part because it lacks evolutionary homologs, eliminating the ability to use animal models for research. Because it is rarely present in normal tissue cells, but often present in cancer cells it is a useful biomarker for the detection of early cancer. NY-ESO-1 has been found in one third to one fourth of all melanoma, lung, esophageal, liver, gastric, prostate, ovarian, or bladder cancers. In cancers including colorectal, pancreatic, renal, and lymphomas, NY-ESO-1 is very rarely identified. The p53 antigen was first discovered in 1979 and recognized as a tumor repressive gene in 1989. It encodes a protein that monitors the cell cycle by suppressing tumor growth and preventing genome mutation. When p53 is defective, abnormal cell proliferation can occur. Mutations in p53 are present in up to 50% of all human tumors. The three major functions of p53 include growth arrest, DNA repair, and apoptosis. P53 halts the growth arrest function of the cell cycle progress which can activate transcription of proteins that repair DNA. There are low levels of p53 in normal cells, but damage to DNA and similar stress signals may precipitate an increase of p53 protein. Cell death is the final course of action to prevent cells containing abnormal DNA from dividing. Term Description
Screening for lung cancer using computerized tomography (CT) has been shown to decrease mortality by 20%. CT is an expensive procedure that also can reveal non-malignant changes on the chest CT. The EarlyCDT-Lung assay is a cheaper alternative with fairly good sensitivity and specificity for detecting early, asymptomatic non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC). To achieve the desired specificity of 90%, the autoantibody cutoff values were chosen based on that goal. The overall sensitivity/specificity of the assay was determined to be 38%/88%. When the sensitivity and specificity were measured based on the type of tumor, NSCLC was 34%/88% and SCLC was 50%/88%.[http://cancerpreventionresearch.aacrjournals.org/content/4/7/1126.long#ref-1]
